in silico pcr (Read 2342 times)

I'm trying to use the in silico pcr tool.  I have primers that match my sequence, with restriction site added to the 5p end.

If I do in silico pcr with only the 3' part of the primer that matches the target sequence, I get a product.

If I add the restriction site to my primer, and set 'mismatches' equal to the number of added bases, I get no product. 

Seems like this should work.  Am I doing something wrong?

Hello! There is a known issue about PCR In Silico (see UGENE-6136). It seems that you have the same problem. If you would like to be sure that it is so, please share some test data with us and describe the steps you do in more details.

Note that the mentioned issue will be fixed as soon as possible, the fix will be included into UGENE 1.31. version.
